چکیده

Cell-free extracts of Rhodococcus corallinus NRRL B-15444R dechlorinated deethylsimazine (2-chloro-4-ethylamino-l,3,5-triazine-6-amine) to N-ethylammeline (2-amino-4-ethylamino-l,3,5-triazine-6(SH)-one) anaerobically, and, thus, presumably hydrolytically. Two enzyme fractions (M, about 450000 and 180000) that dechlorinated deethylsimazine were detected after gel permeation chromatography of cell extracts. trons is not complete [9] and hydrolytic removal of substituents is known [10-12]. We have demonstrated biodegradation of CEAT, involving dechlorination and deamination to EOOT in the coryneform bacterium R. corallinus [13], and we now confirm that the initial reaction is a quantitative, hydrolytic, ring dechlorination by two isofunctional, but different hydrolases.

Deet hylsimazine: Bacterial Dechlorination, Deamination, and Complete Degradation

Enrichment cultures utilizing deethylsimazine (6-chloro-N-ethyl-l,3,5-triazine-2,4-diamine) as a sole nitrogen source were obtained from so5 that had had long exposure to s-triazine herbicides. A bacterium was isolated that utilized deethylsimazine quantitatively as a nitrogen source for growth, but only 1 mol of nitrogen was obtained/mol of deethylsimazine, whereas all six atoms in melamine we...

Biodegradation of Chlorinated s-Triazines

The catabolism of chlorinated s-triazines, e.g., herbicides, is incompletely understood. The first demonstration of their quantitative microbial degradation is the fungal dealkylation of simazine (CEET [for meaning of abbreviations throughout, see Table 1, footnote a]) to two major products, deethylsimazine (CEAT; two-thirds of total) and an unidentified s-triazine (one-third of total) (11, 12)...

Degradation of trichloroethene by a linear-plasmid-encoded alkene monooxygenase in Rhodococcus corallinus (Nocardia corallina) B-276.

Rhodococcus corallinus (formerly Nocardia corallina) B-276, isolated with propene as sole carbon and energy source, is able to oxidize trichloroethene (TCE). Glucose- or propene-grown R. corallinus B-276 cells exhibited no difference in TCE degradation efficiency. TCE degradation was found to be growth-phase-dependent and maximum rates were monitored with stationary-phase cells. K(m) and Vmax v...

Complete dechlorination of tetrachlorohydroquinone by cell extracts of pentachlorophenol-induced Rhodococcus chlorophenolicus.

In this paper we describe the sequence of reactions leading from tetrachloro-para-hydroquinone to 1,2,4-trihydroxybenzene by inducible enzymes of Rhodococcus chlorophenolicus. Tetrachlorohydroquinone was first converted to a dichlorotrihydroxybenzene in a reaction involving both hydrolytic and reductive dechlorination; no trichlorinated intermediate was detected. Dichlorotrihydroxybenzene was s...

Expression, purification and kinetic characterization of recombinant benzoate dioxygenase from Rhodococcus ruber UKMP-5M

In this study, benzoate dioxygenase from Rhodococcus ruber UKMP-5M was catalyzed by oxidating the benzene ring to catechol and other derivatives. The benzoate dioxygenase (benA gene) from Rhodococcus ruber UKMP-5M was then expressed, purified, characterized, The benA gene was amplified (642 bp), and the product was cloned into a pGEM-T vector. The recombinant plasmid pGEMT-benA was digested by d...
